This paper covers the use and importance of toxicokinetics in preclinical safety assessment from the viewpoint of a regulator—the Swedish Medical Products Agency. Examples of significant toxicokinetic issues raised in association with recent general marketing applications are provided. Considering toxicological principles earlier in the drug development process would most likely result in better predictions of the risks associated with the use of medicinal products.
