Technical Note: Isolation of Total RNA
A new automated method for isolation of total RNA in a 96-well format is described in a new technical note. The Strategene Absolutely RNA Microprep Kit allows high-throughput isolation of total RNA from small samples of cultured cells. This simple method eliminates toxic phenol—chloroform extractions and time-consuming ethanol precipitations by selectively binding RNA from cell lysate on 96-well silica fiber matrix plates. The RNA isolation protocol, however, requires extensive manual processing including repetitive pipetting, washing, vacuum filtration, and centrifugation steps.
Using Agilent's VWorks Automation Control Software, a script was written for the Bravo Platform to aspirate, dispense, mix, and transfer sample from a source 96-well tissue culture plate to the binding and collection plates of the Absolutely RNA 96 kit; as well as to automate sample wash steps using a vacuum filtration unit accessory on the Bravo Platform.

According to Fluidigm, its new 12.765 Digital Array is a unique IFC that uses a network of integrated channels and valves to divide a mixture of sample and PCR reagents into 765 replicates. The chip is specially designed to quantify target sequences very accurately and to detect low-abundance targets that differ by only a base pair from the wild-type sequence. With conventional assays, these targets may be otherwise difficult to detect in the sample as a whole. Using a Fluidigm Digital Array, the sample is diluted, mixed with PCR reagents, and loaded into the chip for partitioning. This partitioning in effect lowers the background within any one partition by a factor equal to the total number of partitions. After amplification, the target sequences may be quantified by counting the number of positives in relation to the volume of sample loaded.

2-D Protein Extraction Buer

EZ-2 Personal Evaporator with Inert Gas Purge Module
To meet the requirements of the ATEX directives relating to safe removal of highly inflammable solvent vapors, the EZ-2 now can safely remove most organic solvents without forming an explosive mixture of organic vapor and air. The Inert Gas Purge option purges the air from the evaporator using an inert gas before commencing the evaporation process. The inert gas purge unit includes a key switch to lock the system thereby forcing all users to use the facility.
The inert gas purge option also can be useful where an inert atmosphere is required to help protect dry samples from moisture and/or oxygen exposure at the end of the evaporation process. The inert gas purge module controls gases entering the evaporator; therefore, at the end of the evaporation process, instead of releasing the vacuum by letting air into the chamber, inert gas is introduced instead. Applications Bulletin: Acid Chloride Coupling Reaction Using Ph Controlled Addition
A new applications bulletin from Radleys (TDS05S) describes how a leading agrochemical research laboratory used the Lara Controlled Laboratory Reactor (CLR) to perform a difficult acid chloride coupling reaction using pH controlled addition.
In the described reaction, acid chloride is automatically fed over a 3-h period into the Lara CLR together with simultaneous sodium hydroxide addition to control the pH in the region of 6.5. The coupling of the acid chloride to the sodium salt of an oxy anion is a biphasic reaction in which the temperature has to be maintained at 10 °C to maximize yield of the final ester product. Efficient stirring of the coupling reaction is essential as the product forms as a thick slurry.
According to Radleys, the experimental data demonstrate that the Lara CLR is well suited for automating chemistry of this complex nature where pH control, reaction viscosity, and isothermal temperature control were all challenges to maximizing yield. With minimal training the users were quickly able to set up the system, get the unit running and reproducibly perform the acid coupling reaction to generate high yields of the ester product.

A new ultra-micro volume fluorescence lens cell from Starna addresses requirements for conducting clinical/biochemical/pharmaceutical studies by performing UV/Visible spectrofluorometer analysis on smaller volume samples of typically 5 μL or less, providing a practical solution for when samples cannot be prepared in or diluted to larger volumes.
Starna's new Type 19FL ultra-micro cell, which avoids the need for dilution, is considered well suited for providing analysis in volumes typically handled in molecular biology and forensics applications, and for proteomics applications where the sensitivity of the cell is sufficiently high to measure native fluorescence of proteins prepared in microliter quantities. The cell design also allows the majority of the sample to be recovered.
The new ultra-micro cell is about 5% less sensitive when compared with a conventional 10 mm path length cell. According to Starna, however, the difference is much less than the loss in sensitivity encountered when diluting a sample to the required minimum volume (about 300 μL) for full centimeter-square cells, and the lens design offers greater efficiency than that of a standard micro cell of equivalent geometry.

The EchoTherm Models HP51 and HS51 Digital Hot Plates and Digital Stirring Hot Plates are rugged and designed for large samples. The units feature a membrane keyboards and full function liquid crystal displays where all parameters are continuously visible. These units can store two of the user's favorite settings in memory for instant recall and use at any time.
Both tops are 12 in. by 12 in. square. The ceramic top has a 1400-W heater. The aluminum top has a 1200-W heater. Temperatures can be set up to 450 °C on the ceramic topped units and to 400 °C on cast aluminum tops. The units are readable and settable to 1 °C. Accuracy is 1% over the entire temperature range. Temperature control is by PID software and is controlled to +1 °C or °F. Stirrer speeds can be set from 100 to 1500 rpm. The built-in timer can be set to 99 h and is readable to 1 s.

Multiple Experiment Package for FlowSyn
A new optional Multiple Experiment Package integrates the FlowSyn continuous flow reactor with a Gilson FC203B or FC204 fraction collector to produce a very compact, cost-effective system capable of running up to 10 sequential experiments with different reaction times, temperatures, and reagent ratios. Everything, including the fraction collector, is controlled via the FlowSyn user interface, so no separate PC is needed.
Two operating protocols are available. In fractionation mode, reaction products are collected in programmed fractions. In the unique optimization mode, product is collected, during a programmable window, in a single vial. A small aliquot can also be taken during the experiment and delivered into an LC–MS vial where it can be mixed with a quench reagent so that reaction progress can be measured.

According to VirTis, the mechanical components of most freeze dryers can last in excess of 30 years, but controller platforms are typically updated on a regular basis to increase functionality. Using a digital array from Fluidigm, the Fred Hutchinson Cancer Research Center developed a method for the early detection of a common mechanism of resistance on drug treatment for chronic myeloid leukemia. Researchers were able to detect a specific point mutation, which is associated with acquired resistance to the drugs Gleevec (imatinib mesylate), Sprycel (dasatinib), and Tasigna (nilotinib), as much as 100 days earlier than standard tests.
Fluidigm Europe announced that LGC purchased a BioMark System for genetic analysis in its UK laboratories. The system, with its integrated fluidic circuit technology, adds digital PCR to LGC's existing range of capabilities, which includes quantitative PCR, genotyping, gene expression, copy number variation, and absolute quantification for sequencing.
