Abstract
Elemental analysis of single combinatorial chemistry library members bound to polystyrene beads was studied by energy-dispersive X-ray spectroscopy (EDS) on a scanning electron microscope (SEM). The effects of bead geometry and electron beam exposure time on the elemental analysis results were evaluated. Elemental analysis data were highly variable on round beads, but reliable concentration data could be obtained from flattened beads held in silicon wafer bead holders. The duration of exposure time on flattened beads had no effect on elemental concentration data for the compounds studied. EDS analysis of single beads provided results similar to those obtained by combustion analysis. In summary, elemental analysis of single combinatorial chemistry library members by EDS was shown to be a reliable and fast method of analysis of single beads.
