Abstract
Absorption measurements by transmission through weakly transparent media present some difficulties with conventional commercial spectrophotometers when both good sensitivity and high resolution are required. We describe a home-made computerized spectrophotometer which has been designed for absorption measurements through poorly transparent highly light scattering solutions. Emphasis is given to the difficulties inherent in this type of measurement. Examples are presented to illustrate the effects of the sample thickness and of the fluorescence, which can substantially affect the measured absorbance.
