Eksigent Column Chips for cHiPLC-Nanoflex System
Five new chips expand the flexibility and application of the cHiPLC-nanoflex system for high-performance proteomics analysis including biomarker discovery, validation, and verification, and add higher-flow micro cHiPLC columns for peptide separation at increased throughput levels, as well as new phases for the analysis of intact proteins, phosphopeptides, and glycans.
The first two of the new chips are nano LC columns–C4 for the analysis of intact proteins and graphitized carbon for phosphopeptides and glycans. Three others include 200 µm i.d. C18-CL micro cHiPLC columns in 15 cm as well as a new shorter 5 cm length. These chips are run at 1.0 to 3.5 µL per minute flow rates, reducing gradient delay times compared with nano flow HPLC, providing higher sample throughput and improved chromatography.

Immun-Blot Low Fluorescence PVDF Membranes
These new protein-blotting membranes are optimized for fluorescence and multiplex fluorescence applications. The membranes offer high signal-to-background ratio, low autofluorescence, and superior protein retention to maximize blot detection sensitivity and enable downstream quantitation.
Two primary types of protein-blotting membranes are used in Western blotting applications: nitrocellulose and polyvinylidene difluoride (PVDF). Although both types are suitable for colorimetric and chemiluminescent detection, their high autofluorescence and lower signal-to-background ratios are not suitable for fluorescent detection.
Immun-Blot low fluorescence (LF) PVDF membranes are a low-fluorescence alternative specifically designed for fluorescent immunoblotting and detection in the visible light (red/green/blue) spectrum. Immun-Blot LF PVDF membranes produce highly sensitive and quantitative results thanks to signal-to-background ratios that are three times higher than those of nitrocellulose and conventional PVDF.

Precision Melt Supermix is part of Bio-Rad’s suite of high resolution melt (HRM) analysis products, and according to Bio-Rad, it delivers robust HRM performance for sensitive and accurate detection of single nucleotide polymorphisms (SNPs) and CpG methylation for epigenetic studies. The PCR reagent is compatible with all HRM-capable thermal cyclers.
Traditional genotyping methods, such as denaturing high-pressure liquid chromatography and single-strand conformation polymorphism, involve lengthy protocols and require multiple days for completion. Probe-based genotyping can be expensive, especially for large numbers of SNPs. HRM is a simple, rapid, and inexpensive alternative for studying genetic variation.
Precision Melt Supermix is optimized specifically for accurate, robust, and reproducible HRM discrimination. By employing the saturating EvaGreen dye, which is required for detection of class III/IV SNPs, the supermix discriminates all four SNP classes across a broad range of amplicons. This allows for any genomic region to be studied, regardless of amplicon length and GC content. The supermix is also fully compatible with bisulfite-converted DNA and allows for accurate detection of CpG methylation status. Furthermore, Precision Melt Supermix’s room temperature stability allows users to screen thousands of samples using Bio-Rad’s CFX96 Touch or CFX384 Touch real-time PCR systems with the CFX automation system for high-throughput HRM analysis.

Designed to advance translational research in pathology, specifically in biomarker development, this new portfolio offers updated versions of Definiens Tissue Studio, Definiens Developer XD, and the introduction of a novel component, Definiens Image Miner. According to Definiens, the technology in this product suite provides high accuracy in automatically identifying relevant structures of interest and specific morphology in histology, resulting in a detailed phenotypic fingerprint and biomarker expression profile on a cell-by-cell basis. The large set of descriptors can be correlated against therapy response or patient outcome with the goal of identifying new types of biomarkers of predictive or prognostic value.
The new Definiens Image Miner 1 integrates data mining with image analysis to accelerate biomarker research. The data-mining functionality is based on the same cognition network technology used in Definiens’ image analysis solutions. Definiens Image Miner 1 provides productivity increase when profiling tissue probes for clinically or diagnostically relevant differences in morphology or biomarker expression. Development and quality control are supported by links between data points and the visual inspections of associated regions in the tissue.
Definiens Tissue Studio 3 conducts image analysis for quantitative digital pathology. It combines highly detailed quantification of tissue specimens with a straightforward and fast out-of-the-box configuration workflow. New features include an improved detection of regions of interest, classification of nuclear morphology, and an angiogenesis module.

According to GE Healthcare, its new ExoStar Kit provides a high-performance product for the enzymatic removal of unincorporated primers and nucleotides from PCR and sequencing reactions. The kit enables efficient primer digestion with no degradation of target PCR products, allowing almost 100% yield of the desired PCR products. The PCR clean-up product contains two enzymes that are designed to work together, with one tube of Exonuclease 1 at a concentration of 10U µL−1 and one tube of illustra alkaline phosphatase at a concentration of 1U µl−1.

Aeris Core-Shell HPLC/UHPLC Columns
According to Phenomenex, these new high-performance columns are designed for the analysis of proteins and peptides and deliver higher resolution and peak capacities than other bioseparation media, producing sub-2 µm results on any LC instrument. With virtually no bleed, Aeris columns are highly compatible with MS.
Aeris columns are offered in wide- and small-pore configurations. Aeris WIDEPORE 3.6-µm core-shell columns are optimized for the separation of intact proteins and polypeptides and are available in three selectivities: XB-C18, XB-C8, and C4. Because the 3.6-µm core-shell particles produce lower backpressures than traditional media, longer or coupled columns can be used to increase resolving power on both HPLC and UHPLC instruments.
Aeris PEPTIDE 3.6-µm and 1.7-µm columns, designed with a small pore size, are recommended for the separation of low-molecular-weight peptides and for peptide mapping. The novel XB-C18 stationary phase provides the ideal surface chemistry for resolving peptides. Aeris PEPTIDE 3.6-µm particles offer ultra-high performance at backpressures compatible with both HPLC and UHPLC instruments. On UHPLC systems, the 1.7-µm Aeris particles can deliver higher peak capacities than fully porous sub-2 µm columns.

Phenyl-Hexyl Phase for Kinetex Core-Shell HPLC/UHPLC Column Family
According to Phenomenex, its new phenyl-hexyl columns complement the existing Kinetex phases and are considered especially effective for resolving aromatic hydrocarbons such as tricyclic antidepressants and explosives. Like C18, phenyl-hexyl is ideal for a wide range of applications, including the resolution of moderate hydrophobic compounds. The Kinetex Phenyl-Hexyl is offered in 2.6- and 1.7-µm particle sizes. With complementary C18 and phenyl phases, users can screen for optimal resolution of complex sample mixtures.

An extension of the Phenomenex SecurityGuard line, the new ULTRA is considered ideal for virtually any UHPLC, sub-2-µm or core-shell columns including the Phenomenex Kinetex products. SecurityGuard ULTRA protects against the damaging effects of chemical contaminants and microparticulates introduced by the sample, mobile phase, or system, which can negatively affect column lifetime, method sensitivity, quantitation, and peak identification. Ventana received 510(k) clearance from the U.S. Food and Drug Administration for the VENTANA Companion Algorithm HER2 (4B5) for image analysis applications with associated VIRTUOSO software and iScan Coreo Au scanner. Ventana now offers a complete workflow solution for determining HER2 (4B5) expression in breast cancer patients.
The application assists in the detection and semi-quantitative measurement of HER2 (4B5) protein in formalin-fixed, paraffin-embedded normal and neoplastic tissue. Although the pathologist is still the ultimate authority in scoring HER2 (4B5) stains, the image analysis application helps ensure consistency and objectivity in interpretation. When used with the VENTANA PATHWAY anti-HER2 (4B5) rabbit monoclonal primary antibody, it is indicated for use as an aid in the assessment of breast cancer patients for whom HERCEPTIN (trastuzumab) treatment is being considered. The 510(k) clearance covers all components of the workflow including the 4B5 clone, slide stainer, detection systems, software, and scanner.
