Technical Poster: Versatile High-Throughput Automated Compound Management System
According to Agilent, integration of true noncontact acoustic liquid dispensing technology within an Agilent BioCel System can enable a new level of high-throughput automation in a drug discovery environment. This new poster describes the collaboration of Agilent Automation Solutions, EDC Biosystems, and Pfizer Animal Health Division to produce a fully automated compound management solution for screening and developmental assay plates. Automation and throughput requirements demand that workflows account for manipulation of samples in a wide variety of plate formats, such as 384 well, 1536 well, and 3456 well. The objective of the described project was to reduce human manual steps while increasing the capacity and turnaround time of laboratory workflow processes. With this project, the authors describe a protocol to generate 450 compound plates (384 to 384 dispense) per run.

Single-Cell Expression Data Offers Detailed Molecular Understanding of Embryonic Development
According to Fluidigm, scientists at the Genome Institute of Singapore recently generated significant single-cell expression data crucial for a detailed molecular understanding of mammalian development from fertilization to embryo implantation, a process known as the preimplantation period. The knowledge gained has a direct impact on clinical applications in the areas of regenerative medicine and assisted reproduction. This study is the first of its kind to apply single-cell gene expression analysis of many genes to hundreds of cells in a developmental system.
Using the Fluidigm BioMark microfluidic technology and the mouse preimplantation embryo as a model, the scientists were able to study the expression of 48 genes from individual cells and applied this to analyze over 600 individual cells from the 1-cell to the 64-cell stage of preimplantation development. This high-throughput single-cell research methodology provides the ability to detect dynamic patterns in cellular behavior. Findings of the study resolve some of the arguments pertaining to cellular differentiation events and places fibroblast growth factor signaling as the primary event in the later cell fate decisions.

Access Array System Amplicon Tagging Protocol
A new protocol for Fluidigm's Access Array System enables sequencing of amplicon libraries using GS FLX Titanium Series reagents on the 454 GS FLX sequencing system. The Access Array integrated fluidic circuit (IFC) automatically generates emPCR-ready libraries by simultaneously combining 48 samples and 48 primer sets to produce 48 uniquely barcoded samples per chip for approximately 7
per sample.
Fluidigm's Access Array IFC, when used with a 454 Titanium sequencer, can capture up to 24 kb of sequence data per sample or 1.15 MB per IFC.

Zetasizer Nano ZS
Using the Zetasizer Nano ZS, researchers at the University of South Florida's (USF) Department of Physics are able to perform temperature-controlled static and dynamic light scattering (DLS) measurements of protein hydration, protein interactions, and aggregation kinetics in vitro to gain a better understanding of protein aggregation phenomena, including protein crystallization and amyloid fibril growth. The Zetasizer Nano ZS has enabled USF to monitor the effect on the hydration of proteins by salt ions in solution, measuring the hydrodynamic radius of proteins down to plus/minus one-tenth the diameter of a water molecule. Neither chaotropic or cosmotropic salt ions affect overall protein hydration up to salt concentrations of 1 M.
USF also is using data gathered by the Zetasizer Nano ZS to develop models for protein fibril formation, and also has had success in using the Zetasizer Nano ZS to study the nucleation of gold nanoparticles. Unlike static light measurements commonly used to detect nucleation events, DLS separates out the background scattering from small molecular species in gold sols. Therefore, DLS was able to detect nucleation at a significantly earlier stage than has been possible with other approaches.

According to Microsonic Systems, its HENDRIX SM100 can improve fragment-based drug screening quality by resolubilizing precipitated samples back into solution. Recent data shows that by adding the HENDRIX SM100 to the fragment-based drug discovery process, users are able to resolubilize precipitated fragment libraries and consequently recover the originally lost samples. This process both increases the sample concentrations and improves the accuracy of screening results. Using Lateral Ultrasonic Thrust technology to solubilize, thaw, mix, and suspend is proving to be a highly effective tool to increase screening yields.

Multichannel Verification System
The Artel MVS now generates measurements traceable to national and international standards of volumes dispensed to each well in a 384-well plate. Based on ratiometric photometry, MVS users of 96– or 384-well plates are now able to compare liquid handler performance from site to site and from instrument to instrument. The Artel MVS now can evaluate 16– and 24-channel pipettors, and pipettors with 1, 2, 4, 8, 12, 96, and 384 channels; and users of eight-channel dispensers now can se the Artel MVS to perform accurate volume readings with additional dispense pattern options. With an eight-channel plate reader, the Artel MVS now can read a 384-well microtiter plate 4 times faster than before, in just 1 min. The Artel PowerWave, a monochromator-based plate reader, can be used at a wide range of wavelengths, allowing it to operate with multiple applications. Other enhancements include changes to the system's calibrator plate, a new design that allows for more measurements per cuvette in Data Manager version 2.4 and higher, and easier to clean cuvette surfaces. Existing MVS users receive updates to Data Manager at no charge.

A2s Exoscan and FlexScan FTIR systems are battery powered, handheld FTIR spectrometers that are proven for field applications. With these new portable FTIR systems, samples no longer need to be transported back to the lab. According to Dorton, its NQAD Nano Quantity Analyte Detector is a new and completely different kind of aerosol-based detector for HPLC. Based on condensation nucleation (CNLSD) technology, the NQAD can detect subnanogram levels. According to Fluidigm, its new FC1 Cycler can reduce thermal cycling time by a factor of three when compared with the company's current products, and can reduce overall system configuration costs for users conducting high-sample throughput SNP genotyping because fewer thermal cyclers achieve output requirements. Adding the new FC1 Cycler to an existing Fluidigm system can double the number of microfluidic chips processed in a single day. The FC1 Cycler also integrates a self-contained vacuum source and touch screen interface (that offers easy editing of thermal protocols) in a streamlined design that minimizes lab space requirements.
The new FC1 Cycler is fully compatible with Fluidigm's 96.96 and 48.48 Dynamic Array IFCs and its 12.765 Digital Array IFC. The FC1 Cycler, combined with either the Fluidigm EP1 Reader or BioMark real-time polymerase chain reaction system, can complete the entire workflow of the system in a matter of hours with only minutes of hands-on time for loading the chips. ZEN Double-Quenched Probes
According to IDT, its new double-quenched probe increases the accuracy and reliability of 5′ nuclease qPCR experiments. Although traditional probes have 20-30 bases between the dye and quencher, this probe design positions an internal ZEN quencher only 9 bases from the 5′ fluorophore. This shortened distance, particularly when combined with the standard 3′ quencher, significantly decreases background fluorescence and increases sensitivity. The chemical structure of the ZEN quencher stabilizes duplex formation, which allows for its use in previously validated sequences. The improved functionality significantly increases qPCR accuracy and sensitivity when compared with traditional probes.

Turbo Dicer siRNA Generation Kit
According to AMS Biotechnology (AMSBIO), its new Turbo Dicer siRNA Generation Kit enables easy and cost-effective generation of a large number of small interfering RNAs (siRNAs) from full-length target genes. siRNAs are 21– to 23-nucleotide RNA molecules that can cause targeted gene silencing in mammalian cells through RNA interference.
Using an ultra-active form of human recombinant dicer enzyme, a double-stranded RNA-specific endonuclease, it is possible using the kit to cleave more than 95% of dsRNA template into 22 bp siRNAs within 2 h under optimized reaction conditions. In contrast to individual synthetic siRNAs, the Turbo Dicer kit allows quick production of multiple siRNA species against target mRNAs to achieve effective gene silencing with minimal guesswork. Because minimal concentration of any individual siRNA species is generated with the Turbo Dicer Enzyme, off-target effects are not a problem.

Solastra Five-Color Reagent Panel
Solastra is a comprehensive new CE-marked panel of conjugated-antibody cocktails for the characterization of hematolymphoid neoplasia by flow cytometry. These premixed and ready-to-use panels target the appropriate markers as recommended by the 2006 Bethesda International Consensus and are compatible with the WHO Classification of Tumors of Hæmatopoietic and Lymphoid Tissues. Twenty-three markers are combined in seven tubes—two tubes in the B-cell lineage kit, two tubes in the T-cell lineage kit, and three tubes in the myelomonocytic lineage kit—each tube containing up to five relevant antibodies. Kits can be run individually or jointly.
In addition to CD45, which is included in all seven tubes, the markers represented in the B-cell lineage kit are CD5, CD10, CD19, CD20, CD38, Kappa, and Lambda. Tubes in the T-cell lineage kit offer CD2, CD3, CD4, CD5, CD7, CD8, and CD56. The myelomonocytic lineage kit tubes contain CD7, CD11b, CD13, CD14, CD15, CD16, CD33, CD34, CD56, CD117, and HLA-DR. Cell Lab Signaling Cytometry
The Cell Lab SC line of reagents from Beckman Coulter offers a comprehensive set of antibodies for the detection of the phosphorylation state of signaling proteins in cells via flow cytometry. Performing cell-signaling experiments via flow enables the user to generate precise, specific, and quantitative measurements of the signaling response in individual cellular subpopulations. The reagents, which in the initial offering are conjugated to either Alexa Fluor 488 or Alexa Fluor 647, are released in conjunction with a new method that allows analysis of samples in whole blood. Analyzing samples in whole blood minimizes artifacts and variability introduced through sample manipulation and helps preserve the native biology of the cell. According to Beckman, the method has also shown excellent performance when used with bone marrow, cell culture, and purified cell populations.
